When loading cells (human PBMCs) onto the 10X chromium controller there is a trade off between total cells loaded, recovery and theoretical doublet rate. I am using 2 Hashes so my ability - according to the formula 1-(1/n) - [where n = number of hashes] to detect doublets is 50%. Coupled with the fact that doublet detection software is not perfect is there a general research consensus around the upper threshold limit for doublet formation that one should avoid before it negatively impacts the data considerably?
