I would like to check anticancer activity of compound obtained from microbial origin using simple method which is cost effective and primary. If any one have any method could you suggest?
Generally u can use any of cytotxicity assay methods, I agree with Davoodbasha MTT is v simple, inexpensive and good test. we use CCK-8 in our lab to do any anticancer screening.
In addition to the all suggestions , apoptosis assay could be done using propidium iodide so you check the cell cycling also you can decide we which phase of apoptosis it cause early or late ...good luck
You can determine anticancer potential of natural compounds using different method of analysis of cell death and cell proliferarion. Under normal conditions, there is a balance between cell death and cell proliferation. Natural compound can affect processes of programmed cell death and/or the cell cycle.
you can perform firstly cytotoxic effect for the plant extract, and then apoptotic effect using Annexin v-cy3 and you can confirm your results by testing one of the tranductional pathways such caspase-3.
In my opinion, to properly assess anticancer activity in vitro, you need cancer cells, nonmalignant cells, standard anticancer drugs, a simple cytotoxicity test (MTT, SRB, resazurin, etc), and an answer to the question: Can my compound improve the ability of the standard drugs to kill cancer cells without significantly affecting nonmalignant cells from appropriate tissues? http://www.amazon.com/dp/B00MMO25NM/
