I would like to get a list of medium which can be used for the isolation and cultivation of Halophiles. Which one can support the pigmentation of Haloarchaea and bacteria?
you can use normal nutrient agar and trace metal solution with 40-50 ppm salinity. (preferably you can decide the salinity concentration based on your sampling site). Use low percentage agar for the medium.
Various media including both rich (eg nutrient agar, LB agar etc) or minimal salt medium may be used, but the important factor is the supplementation with salt (eg NaCl). Add 1M or above concentration of NaCl in the growth medium and you will be able to isolate pigmented halophiles.
my suggestion is Zobell's Marine agar media, It is used in the culture of heterotrophic marine bacteria, the minerals it contains closely mimicking sea-water, but with the additional nutrients, peptone and yeast extract.
