I would like to generate human moDCs for 6 days from CD14+ monocytes using IL-4+GM-CSF.

At day 6, when I collect the floating cells, should I also consider the attached cells ?

On day 6, when using different TLR agonists to test their effect on DC maturation for 48h, should I keep GM-CSF/IL-4 in culture ?
