Dear researchers,
I have planned to carry out stress treatment under in vitro condition on half strength MS medium supplemented with NaCl and PEG6000. Should I add NaCl and PEG6000 into the medium before setting pH (5.8) or after setting pH? I'm in a great bias. Kindly help me in this regard. Thank you in advance.

If you consider the pH to be a stress factor in regards to salt stress, then i think you should add NaCl after the pH has been set. Although i imagine that the pH will not change very significantly due to other salts in the MS media buffering the pH anyway. If you dont intend to use pH as a stress input, i think you should set the pH after the addition of NaCl and PEG6000
