Hi, I transfected adherent cells with a plasmid containing a Tet-on/off system and GFP. GFP and my protein of interest are separated by a P2A sequence. The cells were selected with puromycin for one week and then cryopreserved. I expanded the cells and cryopreserved multiple batches.

Recently, I treated the cells with 2.5 µg/mL doxycycline for 72 hours, replenishing the doxycycline every 24 hours. However, only 56.8% of the cells are expressing GFP.

Does this mean that only about half of the cells retained the plasmid, even though in previous experiments around 95% of the cells expressed GFP? Should I perform puromycin selection again before adding doxycycline? Has anyone experienced something similar before?
