I just bought MS liquid medium, micro and macronutrient separately. I wont use the whole medium on a single time, so I must open it several times. My question is, should I manipulate it inside the cabinet, usign sterile glassware or can I use it outside? I obviously will sterilize my flasks when they are ready, but I need to know if the stock media can get contaminated? Besides, I have another question... one of the media changed to a light yellow color. is that contamination?

you can manipulate media outside the cabinet then autoclave it properly. If any heat labile substance you want to add in the media then first autoclaves the media without heat labile substance after that in culture hoot you can add the heat labile substances in to the media after subjected to filter sterlization.
You can use media outside sterile zone and autoclave after preperation. We keep solutions on +4oC to decrease contamination.

Interesting point. Keeping liquid media for long time while opening the bottle many times may lead to contamination and or salt precipitation. Try to store the liquid media at the beginning in smaller flasks and use each one at once. Keep them on the 4° C fridge...
What kind of MS medium is that? Ready to use, or concentrated (ex. 10x) and use it to prepare large volume of MS? I believe yours is the later one, becasue you mentioned 'stock medium'.
1. I would open this stock medium in a hood to minimize the posibility of any contamination, although I don't think 'stock medium' is sterilied beforehand by the company.
2. The solution with 'yellowish' color in one bottle is normal, because it contains vitamins and other micronutrients.
3. If you are doing 'selection medium', and need to add some purchased selection agents, and if these purchased agents are already sterilized by the company, then you should open and close these agent containers in a hood, to keep it sterile for future use.
You can make your final media inside or outside the laminar air flow anywhere but prior to use you have to sterilize it in autoclave for 20 minutes
Dont understand, why you did buy liquid MS . I do use the power MS 4.4 g to the liter. If I dont use the wole liter, I keep the rest in minus 20.
I need MS basic medium gor a lot of different preparations adding hormones etc filter sterilized after autoclaving.

yes you can manipulate but after that you have to do proper autoclave...
If you can autoclave your media 20 min at 121 C (under 1, 2 atm) you do not need to use liquid media and also you can save your money.
If you have seriously contamination problem you need to check your cabinet.
