Hello.

I have a viral plasmid which contains a bicistronic region: CMVprmtr-GFP-IRES-PuroR.

I would like to cut out the GFP sequence to get rid of the green fluorescence. In this case, would it be necessary to cut the GFP together with IRES (since there will only one (PuroR) marker left and there won't be a need for the IRES) or just GFP? I mean could this CMV-IRES, going sequentially, influence somehow PuroR expression? or is it a normal practice to leave just one marker in the IRES constructs?

Also, I would like to ask if anybody could advise me some literature on this topic?
