It's quite normal to see that at higher ODs, when using 96-well plates. I've done hundreds of screens in a log-phase plate reader, and I've seen cells are not very well suspended when the culture is closer to stationary phase. If that is your observation, don't worry about it. Just make sure you seal the plate very well. Best

Thanks so much for your answer. We see clumping of cells at the bottom even before the stationary phase especially for S. uvarum cells that tends to clump more.

Do you recommend round bottom 96-wells?

Our plate reader accepts only flat-bottom plates. I personally don't have experience with S. uvarum, but I think as long as you are consistant and use the same reader for all experiments you're fine.

Are able to see the 3 phases (lag, log, stationary) when you plot the reads? If so, don't worry about seeing the clumping cells.
