I have this cell line of mouse Beta-TC6 cells and I was searching the primary literature to find out how best to serum starve these cells for around 16 hours and I cant find anything. Currently we are growing them is DMEM with 15% serum and 1% Penicillin/Streptomycin. The main reason I want to do this is to eliminate EGF like proteins to be able to stimulate EGFR and related family members.
You did not mention why you would like to serum starve Beta-TC6 cells? These cells express SV-40 large T. Therefore, starvation at lower serum concentrations (0-1%) could result in cell death. More information could help address your question.
I want to look at the response to egf and similar growth factors, which are found in serum normally, so not having serum would be ideal to ascertain the response.

I started working with the Beta TC6 cell line but I am struggling to culture these cells.
Since you are using these cells. Could you tell me if you encountered any problems while working with beta tc6 cells? If you succeded on culturing these cells, could you tell how did you grow these cells?
I am very frustrated at the moment, your input would really help me.
Ps: I am following exactly the ATCC protocol, of which I bought their advised cell media with low bicarbonate content and I am culturing on Corning cell binding flasks.
The doubling time on the ATCC protocol is not happening, these cells take up to one month to actually awake from their dorment state, which they appear rounded and very, very tiny.
