I have combined two haploid Saccharomyces cerevisiae cultures (opposite mating types) in YPD and want to select only diploids without using solid media. Each haploid has a different selectable marker. If I grow them in the appropriate liquid double dropout media will the haploids die or will they start to grow when transferred to a complete media? I'd like to avoid going the solid media route.

Double selection will definitely select the diploid, haploid will die. But you will be 100% sure of the homogeneity of the culture only if you start from a single selected clone...
Not sure the haploids will die in double selection, they will rather enter quiescence if the markers are auxotrophies and not antibiotics. However, they will be overwhelmed by diploids and the portion of haploids will be negligible in the media.
But as Dominique said, start from single clone and do double selection media, if the OD increases, then you have diploids!
