I have been having trouble with SDS PAGE--as you can see there are small bumps in 2nd and 7th (2nd and 8th as well) well. This is precasted gel 4-12%--80V for first 10 min and 120V for 80min. I loaded 70ug/ul-- I know it's a lot, but it's because that the signal of gene of interest is very week. I've had no issue with this loading amount when I ran with mouse heart samples. These are human heart samples. I homogenized and sonicated, and did the BCA (duplicated--and two values were pretty consistent). If you have ever seen this or have any solution, please let me know!!

How do your final results look?
That kinda reminds me of some old gels I ran with lots of urea or guanidine in them. If you have large quantities of salts or reducing agents that aren't being diluted out you may want to run them through desalting columns before doing your SDS-PAGE.
5% BME is pretty standard in SDS-PAGE samples, but if you go much higher it could definitely make your gel run a little wonky. Are these different samples or all the same? If their different you may just have different levels of signal. If so your final result looks pretty good so I wouldn't worry about how wonky it looks while running.
