Hello everyone, I was just wondering if someone could offer some advice or point me in the direction of a paper that could answer my question?

We currently have some femur samples that have been collected from pigs, and I wish to do some histological TRAP staining in order to observe osteoclast cell numbers. I'm aware that the general procedure is to fix the tissue and decalcify before any staining is done, but I want to be very careful in how I actually sample the bone before fixing. There are questions such as: what would be the best method to cut a sample from the femur prior to fixing, and should TRAP staining be done on the surface of the cortical bone, or should we aim for staining a cross-section of the bone?

Therefore can I ask if anyone has prepared whole bone samples for histology before, or knows of any good papers that could point me in the right direction?
