Dose anyone generate Knock-in and Knock-out mouse by CRISPR-Cas9 system? I constructed knock-in and knock-out mouse by CRISPR-Cas9 system?should i use southern blot technique to screen? is it necessary to check by Southern Blot except for the PCR and sequencing test? what is the consideration of Southerb Blot screening?

Hi there,
If genomicDNA is significantly affected by the Knock in/Knock out compared to wt (different distances between primers) then PCR on genomic DNA is very straight forward and PCR product might also be purified for confirmation by sequencing.
