please guide me about it,,,
polysaccharides (eg capsule) induce only humoral immunity, and igM will only produce against it,but igG will not,, but if conjugate it with some sort of protein carrier,,so now it will capable to activate cell mediated immunity as well as igG antibody,,,,
dear friend,
Polysacchrides can easily be directly detected by the antibodies, but not as proteins which must be processed and then detected by T cells through MHC complex. hence Polysacchrides are induced only by humoral and proteins by both humoral and cell mediated. Cell mediated immunity does not does not get induced by polysacchrides since Polysacchrides are not processed and bind to MHC complex.
IgM will only produce against it,but igG will not?
since IgM is the first which is produced at the beginning of the infection and it is temporary. It will neutralizes the infection until the IgG is produced and The IgG conjugates with carrier protein inorder to activate the phagocytic cells. or combines with complement system to activate the T cells, mast cells and other B cells for more immune response.
Their is only one action of all antibodies: activation of other immune cells.
you must also note that IgM is pentavalent antibody. Hence it has large diversity to bind any polysacchrides or protein...

the switch of IgM to IgG and other isotypes is a process named class switch recombination (CSR) and it happens in T-cell dependent antibody responses and one from its major sites in the body is the germinal center reaction which happens in lymphoid follicles as a response to T-cell dependent antigens inspite of that CSR can happen also in some T-cell independent responses but to a very less degree and it will also needs some T-cell cytokines.
