I've been extracting microbial RNA from environmental samples (mud) and ran my extraction on the Bioanalyser to check the integrity.

I've obtained surprising results : the sample with a ratio 23S/16S= 0.5 had a RIN of 7 (sample 1 on picture) whereas the one with a ratio 23S/16S= 0 had a RIN of 7.1 (sample 2 on picture), which would indicate higher integrity.

Has anyone experienced such anomaly?

Is it possible that the RIN algorithm fails when 23S/16S= 0?
