If on day 1 i revived the cells and on day 2 its confluent enough to be seeded on 6 well plates. Can i do experiment on the 6 well plates on day 3? Or is it too early?
I did spin down away the dmso during revival of frozen cells
This depends on the cell line, but I would be careful about using them too early. They'll be particularly fragile and may end up giving you misleading results. Recommend giving them a few days minimum for them to purge damaged cells and repair damage from freezing.
I would suggest you to wait at least 2 passages to start the experiment, depending of the cell type (5-7 days). After thawing, cells need time to recover and establish normal proliferation cycle. If cells did not recover properly after thawing, this will greatly influence your future experiments. So, doing the experiment on day 3 after thawing is too early.
