Please check the polartity index chart of solvents i. e. polar, nopnpolar and mid polar solvents and the polarity of β-1, 3-glucan.
TLC and HPTLC does not recommand buffer solutions. you prepare your solution in a solvent and while preparing mobile phase (approx 20ml) for HPTLC Twin trough chamnber add to it one to two drop of acetic acid or ammonia.
Sharad Medhe Thanks for your suggestions, However the B 1,3 Glucan completely dissolve in 1M NaoH solution, So, could I see the band if I add acetic acid or ammonia in mobile phase (in chamber) ?
1M solution of NaOH is highly strong base. It will useful to move on TLC or HPTLC plate. You check the solubility of B 1,3 Glucan as in Polar, Non Polar or either midpolar solvents. then u chose the mobile phase and according to the seperated band quality you may add acetic acid or ammonia in mobile phase (Prepare it in 100ml Conical flask and after mixing pour it in TTC tank).
