While redox potential is an important factor, it is not a standard measurement routinely monitored in cell culture. Data sheets for cell culture media like DMEM typically provide information on the components, pH, osmolality, sterility, and other quality control parameters. Redox potential, being a property of the solution rather than the medium itself, just as a characteristic of the solution formed when the medium is mixed with other components like serum, is not usually included in the data sheet. This is the reason why a specific redox potential value for a 10% FBS-supplemented medium isn't readily available.

But the culture medium supplemented with 10%FBS will generally be maintained in a reducing state due to the presence of FBS and other components that contribute to a more reducing environment. However, this might not always be true. While FBS can contribute to a reducing environment, it is not the only factor determining the overall redox potential. If the culture medium is exposed to atmospheric oxygen, or if other oxidizing agents are present, the redox potential may not be significantly reduced.

You can always measure the redox potential of DMEM supplemented with 10% FBS using an electrochemical probe. An electrochemical probe (like a potentiometric sensor or a redox electrode) may be used to directly measure the redox potential of the solution (DMEM +10% FBS). This involves applying a voltage and measuring the potential difference.

Redox potential (Eh) of DMEM + 10% FBS under “standard” cell-culture conditions is not a single fixed number: measured Eh values depend on medium composition, dissolved oxygen, CO₂ incubator conditions, temperature, and cell metabolic activity; reported baseline Eh for typical cell-culture media equilibrated with atmospheric/CO₂ conditions often falls in the mildly reducing to near-neutral range and can shift by tens to hundreds of millivolts with cell respiration — therefore measure directly with a calibrated redox electrode under your exact protocols rather than rely on a single literature value.
