I am planning an experiment using TTX on C2C12 myotubes. The TTX should be reconstituted in citrate buffer pH4.8. I am now trying to find out which molarity of citrate buffer can be used for C2C12 cells. I am reading different things from 1.5mM to 100mM. My feeling is that the lesser the better.
Has anyone used citrate buffer in C2C12 myotubes and has a recommendation for the molarity?

The citrate buffer molarity probably doesn't matter all that much as long as the media pH is not affected and media osmolality is maintained between 260 and 320 mOsm/kg. Assuming you will be using submicromolar levels of TTX in the media, your TTX stock solution can be quite concentrated. In that case, you could probably use 100 mM citrate buffer for your stock solution and have no problems.
