I have been using Basement Membrane Matrix Gel to do cell invasion experiments with transfected cells, and now I want to add a drug X (X can promote cell death) to observe its effect on invasion.

I have a few questions:

1. For drug X that induces cell death, should I add it to the upper chamber, the lower chamber, or both?

2. I also want to study how the expression of gene A affects the action of drug X and cell behavior. To this end, I plan to use drug X and inhibitors/agonists of gene A (small molecule modulators) at the same time. Can X and small molecule modulators be added at the same time? If so, should they be added to the upper chamber, the lower chamber, or both?

Or, is it better to add the small molecule modulator first and then the drug? If so, where should they be added?
