Hello, I would like to ask if there is a significant difference in the size of genomic DNA obtained by different bacterial genome extraction methods (such as mild enzymatic lysis or violent physical disruption methods)? Recently, I used a glass bead disruption method to extract genomic DNA from a strain of Streptomyces. The final DNA obtained after agarose electrophoresis analysis always had a slightly lower band than the 23 kb marker band. Is this normal? If I use a gentle genomic DNA extraction method, can I obtain more complete genomic DNA? Thank you for your attention and reply.
