I have been attempting to purify a streptagII-tagged protein from mammalian cells using streptavidin-magnetic beads without success. The reason I'm using streptavidin and not streptactin is that there is evidence showing bead diameter is an important factor in purifying for my purposes and streptatin beads are not available in the size I need.

I've tried lysing cells without and with detergent (1% triton x100) but I have not been able to recover my tagged protein or binding partners under either condition.

I see a number of posts about streptagII but I dont see any examples of folks getting this kind of assay to work. Does anyone have experience with this?
