I am doing synthesis of Octapeptin C4 analogues with variable tail moieties as part of my project. I have made the final compound, which is cyclized and all BOCs removed. The problem is, I can't purify it in GILSON PLC-2020 using XDB-Phenyl column and 0.1% TFA in water/acetonitrile we have in Cooper group since it gives trouble sometimes. So, I tried to purify on GRACE Reveleris using water/acetonitrile. But I also failed to get it on GRACE. Surprisingly, I was expecting that using a C18 in GRACE would give me the desired product which has a retention time of 3.87 min, confirmed by LC/MS on C18 column.
Can anyone please suggest me an alternative way to get the product? I don't understand why GRACE is not giving me the pure product whereas I can see it on LC/MS using C18 column.
