Dear Colleagues

I tried to concentrate protein from yeast. Briefly, I grew yeast in YEPD media, centrifuged, and the culture supernatant was filtered. The clarified extract was precipitated with ice cold ethanol to a final concentration of 70%(vol/vol). After centrifugation the pellet obtained was re-suspended in citrate phosphate buffer pH 4 and activity was tested against other closely related yeasts. I did not get activity. But, when I Freeze-dried the clarified extract and did activity assay, got activity against other yeast species. 

Does anybody have suggestions how can I precipitate protein from my sample with activity?
