Hello colleagues, I would like to manipulate female mice to induce pseudo-pregnancy by injection of Prolactin. Is anyone familiar with this procedure because I have no idea if I would need to inject Prolactin several constitutive days at which dose to really induce pregnancy-induced mammary gland development? Furthermore, does anyone know how far Prolactin goes OR let me ask it this way: Will Prolactin induce the pregnancy-dependent mammary gland development until the step of milk production or will it stop at an earlier stage of gestation? I do something about transgenic mouse, and I also use the pseudopregnant female mice. So my advise is that you can buy some sterilized male mice from commercial animal centers like JAX mouse and then you can cage the erotogenic female mice with sterilized male mice together ahead of a day to obtain pseudopregnant mice the next morning.

Thanks to both of you! Although, my approach is to analyze the pregnancy-induced mammary gland development in the adult female mouse after knockout of my gene of interest. Since I am using a Tamoxifen-inducible conditional mouse model I need to inject Tamoxifen into the adult female mice to excise my gene of interest. Afterwards, I am waiting two to three weeks to breed my mice to induce pregnancy-associated mammary gland development. Since Tamoxifen is an estrogen derivate the remaining not metabolized amount in the female mouse is preventing the normal implantation process of the egg in the utero of the mouse. Therefore, I thought of using an hormon to induce just the pregnancy-associated development because I do not need the pups finally (they are dieing in my knockout mice anyway due to lactation failure). Can I then just go via sterilized mice anyway? I would need to induce the milk production process in the female as well as the so called lobuloalveolar development. Thanks again, Nicole
Mehvisk, I have no idea what the developmental stage of the mammary glands of mice at the end of pseudo-pregnancy is. How far the glands are developing and how far the pregnancy stage is going in the case of pseudo-pregnancy. That is something I was going to hope to get an answer for. Thanks, Nicole
