I work on potato tissue culture and have problem with low regeneration rate. I know using TDZ and Zeatin this problem can be solved but these two materials are so expensive and not economical. So, can anyone help and provide me a protocol without TDZ and Zeatin with high frequency regeneration?
You got already shoots?
If you have leaves try MS plud 1ppm BA plus 0.1 ppm IAA in dishes.
You can use one of the alternatives for these two substances and try them on the food medium used with some plant extracts

I would suggest to start with optimal culture medium with optimal nutrients ratio, what is much more effective to compare with playing with hormones.
For the shoots you have to induce internal auxin production and create stem cell niche. Cytokinin is a good tool for this. But do not forget that each nutrient component can increase or decrease cytokinin effect. Medium must be well-balanced specifically for shoot induction!
