After 3days of splitting I am facing turbidity problem in L929 cells(Fibroblasts),But there are no symptoms of bacterial contamination.. Some protocol says to isolate conditioned medium from L929 cells, they will keep for 7 days..if that is true..then is this turbidity due to release of metabolites? Please suggest me, what could be the reason?
I have made many turbidity measurements in various types of surface waters during my career. There are various types of turbidity meters used during my career, but for surface waters, the Nephelometer type is the standard. It sends its light up from bottom of bottle and measures light refracted in sensor. It can be affected by not diluting when turbidity exceeds 40 NTUs or if particles settle or accumulate in bottom, masking light entry. Mineral (sand, silt or clay) as well as organic particles can reflect light and be measured as turbidity. For water, the standard typically used NTUs (nephelometric turbidity units - forgive my spelling) is formalin (again spelling?), and this may not be suitable standard for the type of particles used in your work which I am unfamiliar with. My point is to understand the turbidity meter, now it works, understand your particles, and standards for reference and dilution in Standard Methods for Analysis of Water and Wastewater (if greater than a turbidity of 40 NTUs).
The turbidity techniques may be slightly different in your profession, so you may have to consider that in your analysis of potential contaminants or conditions that affect results.
