Hi all, I am doing Forskolin Induced Swelling (FIS) assay. It used to work very much fine a few months ago and all of sudden it has stopped working (or instead of swelling it shrinks now !). I have tried different variations, started the assay with all new reagents but not helped yet.

We do FIS with lung organoids in 25% (or 50%) Matrigel with 5 micro Molar Forskolin final concentration.

Any suggestions would be highly appreciated !!!
