Recently, I used different ethanol concentration to treat cells. The ethanol is dissolved in the DMEM cell culture medium. But I found one problem is that the ethanol will evaporate from cell culture medium. I used the small well plate to culture the cell. I was thinking how to prevent the ethanol evaporation. I searched several papers they suggest to create a saturation environment of ethanol to prevent the ethanol evaporation. I wondering high concentration ethanol atmosphere will affect my experiment results. Also, sealing the well of well plate will affect the O2 level to cells. After searching the materials, I found the silicone oil (Anti-evaporation oil). Is it possible to prevent the ethanol lossing from cell culture medium, by cover the cell culture medium with a layer of these oil.
