I did cell cycle assy with PI stained cells using FACS analysis and found my drug has the G1 block effect on cell cycle. The percentange of G1 phase increased from 63% to 78% and correspondingly the percentange of S phase decreased from 13% to 5% and the G2-M phase also decreased form 24% to 16%.So my question is what's the possible reason for the potential G1 phase block? And what kind of experiment I can do to corroborate this?

I do not know enough about the cells you use. But you may have a p53-dependent cell cycle arrest. Your drug may induce DNA damage and trigger this response.

yes, check p53 and also look for the phosphorylation status of pRB, and G1 cyclins. 3D telomere organization also has a specific G1 signature.
