I have a question regarding the phenotypic stress assay. If I am conducting salt stress testing on three genotypes (including WT) under different salt concentrations ( e.g: 50mM, 100mM and 150mM) , how many plates do you typically use per replicate? I am considering using 30 seeds per genotype in each repeat. Does this approach sound appropriate?

When deciding to measure root length, under which conditions and at what concentration should I expect to see changes before making a decision?

Looking forward to your advice.
