The question is not clear. First of all which petroleum you will use? I guess diesel or crude oil probably. Here some comments for you:
1. In bacterial culture for biodegradation purpose (i guess this is your purpose), the solubility of compound is a factor. I assume that you may give more petroleum than its solubility in water. In that case, common practice is to shaking well on rotary shaker. But for comparison with the time of degradation (e.g., time 0 and so on), I would suggest a destructive sampling method, which will reduce the bias sampling from the liquid culture as the solubility is a problem.
2. GC-FID for you desire petroleum, follow one or two good journal articles-often details are written there including the column you need. Standard you have to buy from supplier, such as Sigma/Superclo etc.
3. For FTIR, I doubt about the quantitative analysis without any reference material. Even though you run reference material, I would suggest to take FTIR results as the semi-quantitative or qualitative evidence whether your compound (petroleum) removed completely or not. Thanks

What I need to know is how we can analayze reseidual petroleum by GC-FID,
In general, when we run a GC-FID, we do with standards, here in my case I want to quantify the residual petroleum in my bacterial culture in comparaison with the control and time 0.
To do that, I will compare the peaks in each treatment, So what should I take on consideration to do that ? adn Can I know the peteoleum fractions ( alkanes and aromatics) from this technique or no ??
