I have been working on Anthelminthic drug resistance in nematodes parasites. But currently I am struggling to get pcr results as I am using isotype _1 Beta tubulin Primers for illumina Miseq and deep amplicon sequencing to be done but my 1st round pcr did not work? while the same protocol does work with one of my colleague now I am repeating the same technique for mixed nematodes L3 larvae and even my positive and negative sample did not work. while I am using KAPA enzyme! its might be handling or pipetting problem ? or there would be some other problem ? I am desperately waiting for appropriate answer to my question ?
