Thank you for the comments. I have used in my previous lab Tween 20 at 0.1%, and in my current lab all use Triton X100 at 0.05%. Both were working well without problems. But, when I used an antibody from my previous lab with TritonX I didn't see protein bands. So, I wanted to know if Triton is the problem, or any thing else.

I think you should use 0.05% Tween-20 in PBS (PBST). it is one of the most used recipe and works for almost all western blots.

we are using TBST(50mM Tris pH 7.5, 150mM NaCl, 0.05% tween -20) for regular westerns. In same buffer we are making blocking solution.

I have also used Tween (although it was 0.1%)

Yes, both will work. Both are detergent where Triton X100 is stronger than Tween 20 and need less amount. Triton X100 will be problem if you use more.

0.1% Tween-20 in PBS or TBS  works great for western blot. 

 0.1% Tween-20 in PBS. It works well. 

Hi Vinoth...can I know if your problem was sorted? Was ur protein band not coming because of Triton X? And what % of triton x 100 worked good? I tried 0.1%, but my whole blot was glowing. 

Triton-X absorbs UV (and fluoresces) and should not be used if you are using certain immunofluorescent stains.
