We cocultured human epidermal keratinocytes with allogeneic PBMC in Human Keratinocyte Growth Supplement (HKGS), Epilife medium, Dulbecco's Modified Eagle Medium (DMEM) with autologous serum (with Peripheral blood mononuclear cell (PBMC) at 20% final concentration and standard antibiotic/antimycotic.

Today we saw that somehow the PBMCs were clustered and clumping on random locations on all wells..none of the seem to be attached while those PBMC not cocultured but using the same mixture of medium were attached and do not cluster.

Is this some kind of Lymphocyte reaction?, I am not well versed in immunology so can anybody give me some insight?

The experiment was designed according to Alogeneic Mixed Lymphosite Reaction (MLR) which is new to me.
