We have been using hPBMC-NOG mice for humanized mice. After tumor has grown to a 800mm3, we have done immunophenotyping of these tumors. We consistently observing that CD4+ cells are completely lost and only CD8 cells are there in CD3 gated cells. Is some thing anybody else also observed? Any suggestions or explanations will be appreciated.

I am not sure of the above observation, but more information would help others to answer your question. The most important factors include the duration from PBMC inoculation to the analysis. Hu-PBMC can be only used for short-term studies (< 3 months). If you did not analyze peripheral blood mononuclear cells or splenocytes, then do that and compare those data to the above data on tumor-infiltrating T cells. You may also clarify the method for the above-mentioned immunophenotyping.
