Hello I am new in the nanoparticles research field. My master project includes the development of a nonextruded LysoPS:DMPC nanoparticle loaded with an antibody. What will you recommend to get small size particles (around 200 nm) without extrusion?
We use the thin film hydration method to produce the liposomes.
Any suggestion that what might I try will be highly appreciated.
I've been reading about the different conditions that affect liposomes size and encapsulation efficiency.

I made these tables this year, in there apparently people prepare liposomes using only sonication for size reduction. I have never tried that. Moreover as you can see in the tables; proteins are mostly adsorbed after liposome preparation.
After thin-film hydration people apparently uses ultrasonication for size reduction. If an antibody is added I presume it will be destroyed by the treatment, hence my comment to first prepare the liposomes an then adsorb the antibody. If the antibody must be trapped in the liposome, then extrusion must be followed.
