I have tried many times for PCR of Citrus canker pathogen (Xanthomonas axonopodis pv.citri) but couldn't find the results. I also carried experiment both on extracted DNA of bacterium and of infected tissues. Even though, I also used gradient PCR from 50 C to 62 C (annealing temperature) a couple of times. I used the following primers.

I used the following recipe 

Desired DNA=(1, 2, 3 μl) 

Primer = ( 2 μl) diluted 1.5 μl back and 1.5 μl forward in 97 μl water.

MgCl2  = 2μl

Taq Buffer = 2 μl

Taq Ploymerase = 0.2 μl.

dNTPs = 2 μl.
