I am conducting experiments where I want to assess the glucose uptake (using 2-NBDG) in unstimulated, M1 and M2 macrophages. What I currently do is I seed the cells and polarize them towards M1 and M2 phenotype. I starve the cells (-glucose, - FBS) for 2h prior to incubation with 2-NBDG (30 mins, 100uM). I do not see a difference in glucose uptake between the conditions. I expect a difference in glucose uptake, since M1 macrophages rely primarily on glycolysis and M2 on OXPHOS.

Does anyone has a suggestion for a protocol change or something else?
