I am currently working with VEGF PLGA nanoparticles for lymphedema treatment. I am using PVA 1% as a stabilizer and dissolving 0.1gm PLGA in 4ml DCM. I am following multi emulsion (w1/o/w2) by sonication --evaporation method. Finally lyophilization is done for 48h to form nanoparticles. But unfortunately, in my case the product so formed after lyohilization is soft, cotton texture rather than nanoparticles. I made an attempt by changing the PVA as well as PLGA concentration. But the nanoparticle size is quite large around 1 micrometer and the result is not reproducible. So, I am in need of your valuable suggestions.
Have you tried threalose 5% during the freeze-drying process??? Probably you need a cryoprotectant
