I treated them with 70% alc, 2.5% NaOCl, 0.1 HgCl2, and 1g/l bevastine. These chem were used in diff combinations and for diff time periods. Infection still occurs. Please suggest any other way of disinfection and sterilization.
Was the forceps you used to move the sterilized seeds to medium sterilized too? We autoclave our forceps beforehand. Also, we dip it into 70% ethanol, flame it briefly before touching sterilized seeds (after cooling down from the flame). Also, was your medium sterilized properly? The autoclave worked 100% properly?
Wipe the stems of tobacco with 96% ethyl alcohol and then additionally sterilize with a 0.11% aqueous solution of sublimate. Pieces of the stem are immersed in a solution of sublimate for 15 minutes. Tobacco stalks are washed with sterile water from sublimate. In the last glass, the stems are left and taken periodically for work.
Yuan-Yeu Yau The forceps were sterilized properly and the medium was properly autoclaved. The issue is that the seeds are infected with fungus and usual surface sterilization is not working.
Fresh, healthy seeds generally do not get infection and even do not require any surface sterilization. However, in certain cases especially stored or old seeds, due to loss of viability/dead tissue and/or storage condition, the seeds get infected within the seed coat and can not be sterilized by usual surface sterilization.
