I would like to ask you about MIC first, after I searching in the internet about my query I could not found any answer. My question is about how can I calculate the concentration of stock solution? For example, when I do MIC by macrodilution method I make serial dilution of 10 tubes, then, I 1 add ml of M&H broth in each tube, and I added I ml of 30mg\ml of my drug. So, the concentration of the first tube will be 15mg\ml and so on.

If I add 1 ml of 0.5 McFarland of the bacterial growth in normal saline to get 5x105. Is the concentration will be changed after I added 1 ml of bacteria? I.e the concentration should be divided by 3 not 2? For example, tube no 1 conc will be 10mg\ml not 15???
