Dr. Narayanan,

You can consider disrupting the matrigel in order to dissolve the formazon crystals with acidified isopropanol instead of DMSO.

Hi Narayanan, you could use trypsin and/or proteinase K to hydrolyze the Matrigel, spin out the cells and then do the MTT assay.
