My class is isolating antibiotic-producing bacteria from soil. The majority of the isolates are Bacillus species since we use nonselective media (grown on trypticase soy agar, LB, or potato dextrose agar). We also get Pseudomonas species on this media. When we use phenylethyl alcohol (PEA) or MacConkey agar, we do not isolate very many colonies. I would like to look at Gram-positive bacteria other than Bacillus species. Does anyone know of a way to reduce sporulation and recovery of Bacillus and enhance recovery of non-Bacillus species? Most of the treatments to reduce spores will kill all of the bacteria. Any ideas?
