I would like to measure the intratesticular amount of testosterone in hamster (rodent). I have seen some articles using a RIA approach on testicular tissue homogenate.
Could anyone give me more information about this approach, any alternative and mainly instructions on how to do the testis homogenates and kits/methods for the RIA or other techniques?

Tissue is centrifuged at 2271 g in 2 ml of 0.1 M phosphate buffer containing 0.01% thimerosal for 15 min; the supernate is centrifuged again at 13500 g for 15 min to remove any remaining unhomogenized tissue. The testosterone is commonly measured by ELISA.
