Has anyone tried measuring contractility in hiPSC derived cardiomyocytes? If yes, what method and machine is required to measure the contractility vs time?
To summarize the protocol, we create patterns by employing soft lithography techniques and measure the contractility using videos captured on a stereomicroscope. We used custom-MATLAB codes to measure the stress but the reference provided has all the relevant equations used. The thin films were created using PDMS elastomers but based on the availability of resources, you can also create gelatin thin films.
Very easy to measure with this method, see paper“MUSCLEMOTION: A Versatile Open Software Tool to Quantify Cardiomyocyte and Cardiac Muscle Contraction In Vitro and In Vivo” fulltext link:https://www.ahajournals.org/doi/10.1161/CIRCRESAHA.117.312067?url_ver=Z39.88-2003&rfr_id=ori:rid:crossref.org&rfr_dat=cr_pub%20%200pubmed
