I have a 231 cell line growing but it the media is getting cloudy periodically. I have changed the media, PBS, trypsin but still it is getting contaminated.
I thought it might be overgrowth but I diluted the split to 1:10 but still getting contamination aka cloudiness. I am not sure what is the issue here.

Cloudy media can be bacterial contamination. Do you routinely check your media without cells for sterility? When you use or make the media, do a 48-72hr check by placing some in a flask in the incubator first. When that is clear then you can use the new media with the cells. You should have an ongoing stock and new stock. Same goes for cells, you should have back up flasks and frozen cells.

I have tried all the steps mentioned above but I am still getting contamination and cloudy media.
Christine Pacula-Cox I have checked media by it self the media is clean and no contamination. But with the cells it is contaminated.
When I thaw a new vial of cells they are fine but after 2 or 3 passages the media becomes cloudy. I am very confused I cleaned the incubator and the hood with bleach and 70% ethanol. I dont know what to do now.
The cell line it self might not be contaminated as I have been using this batch of cells and I have been using this for a year they were fine but now I am getting this contamination for the last few months.
Any help is appreciated.
