Hello everyone, I am trying to use Popgene 32 software for analysis of binary data for dominant marker of five ISSR primers across five populations and having 38 loci. Unfortunately the program gives me error by uploading the notepad file, however I have followed the method of entering data.

Hi, It's been a long time since I don't use Popgene, but I guess probably your parameters might have to be corrected. I think you need to set number of populations = 5, and number of loci = 38. The number five that you mention as primers might not be needed. Then for each individual (each row) you will have 38 data (either 0 or 1).

You need not to upload your data prepared in a note pad. You just open a dominant marker file provided with the POPGENE 32 (named as RAPD) and modified the information on that by pasting your data (if you use ctr+V function it wont work, just click right side in the page and paste it). Spacing between each parameters and information is very important in this program. If it vary the program will give you error. Save that file using save as function. Once saved close the file in program and reopen your data and analyze it. It will definitely work, if not txt again.
