Hello everybody,
Does someone have any experience with assessing/extimating mitochondrial activity on FFPE tissues? Precisely, no TOM20, ATP5B, etc: we are trying to get an approximative idea of the cellular metabolism and not of the number/morphology of mitochondria. Something similar to the mitotrackers for cell imaging or a IHC scoring system or a particular type of chemical staining may all work.
I feell this might already be a known problem for this research field, but I hope to find someone who knows better than me. Negative answers are also welcomed!

I have not worked in this area but I can help you out in some way. For assessing mitochondrial activity on FFPE tissue you can try the following markers for which antibodies are available.
Cytochrome c
HSP 60
Hexokinase-1(HXK-1)
Succinate dehydrogenase (SDH)

Malcolm Nobre Thank you for your suggestion. Unfortunately these markers are very similar to the ones I cited and, although they mitochondrial markers, they are rather gross in extimating the actual mitochondrial activity, giving only an extimantion of their number or of the expression of the protein.
Something I found to improve the reliability of these markers is a so called BEC index, which indeed takes into account indeed HSP60, ATP5B and GDPH. However I am not sure of how much this score is validated/reliable either...
Also very interested in any suggestions here! Would dyes like Mitotracker CMXRos not be retained in tissues after fixation/paraffin embedding?
